Abstract

Background: Thioacetamide causes kidney dysfunction. The present study was done to evaluate the protect effects of fish oil omega3 Supplements and aqueous extract of Glycyrrhiza glabra root on disorder of renal function induced by thioacetamide in male rats.

Materials and Methods: 63 male rats were divided into 9 groups of 7 : The control group, The sham group receiving 0. 4ml/kg olive oil as the solvent of Fish oil Omega-3 supplements, the thioacetamide group interperitonealy receiving a dose of 150 mg/kg thioacetamide at the end of the experiment, the experimental groups 1, 2 and 3 orally receiving a daily dose of 100, 200 and 300 mg/kg fish oil omega-3 supplements respectively for 3 month followed by an inter peritoneal dose of 150 mg/kg thioacetamide at the end of the experiment. experimental groups 4 and 5 and 6: they received the aqueous extract of Glycyrrhiza glabra root at the doses of 100,200,300mg/kg daily orally during 3 months respectively and then a single dose of thioacetamide at 150 mg/kg as interperitoneal injection. The levels of serum creatinine, BUN, sodium and potassium were measured. The pathological changes of tissue samples of the kidneys were studied after hematoxylin-eosin staining.

Results: The mean serum levels of potassium in the second experimental group significantly decreased compared to the group receiving thioacetamide . The mean serum sodium in all experimental groups decreased significantly compared to the group receiving thioacetamide. No significant changes were observed in the mean serum levels of FBS, BUN and creatinine in all experimental groups compared to the group receiving thioacetamide. Pretreatment with the aqueous extract of Glycyrrhiza glabra root at all doses let to increased the serum level of BUN comparing with thioacetamide group but there was no significant difference . Pretreatment with the aqueous extract of Glycyrrhiza glabra root at all doses let to significant reduced the serum levels of potassium and sodium comparing with thioacetamide group. Pretreatment with 300 mg/kg of aqueous extract of Glycyrrhiza glabra root let to significant reduced the serum level of creatinine comparing with thioacetamide group. All the experimental groups improved renal histological changes induced by thioacetamide and these protective effects were dose-dependent. (p≤0/05)

Coclusion: The results of this study showed the protective effect of omega-3 fish oil supplement and aqueous extract of Glycyrrhiza glabra root on disorder of renal function induced by thioacetamide in male rats.

Abstract

Background: Thioacetamide causes disorder of lipid function . The present study was done to evaluate the protect effects omega3 fish oil supplements on disorder of lipid function induced by thioacetamide in male rats.           

Material and Methods: 42 male rats were divided into 6 groups: Control group, Sham group . Animals of this group received 0/4ml olive oil as a solvent omega3 fish oil supplements orally per day for 3 months. Thioacetamide group. Animals of this group received150 mg/kg of TAA intraperitonealy in a single dose for 3 months. Experimental groups 1,2 and 3. Animals of this groups received 100,200 and 300 mg/kg of omega3 fish oil supplements orally per day for 3 months and 150 mg/kg of TAA intraperitonealy in a single dose for 3months. All animals bled after 3 months. The provided blood samples were tested for total cholesterol, LDL and HDL cholesterol, triglyceride and glucose serum levels by enzymatic methods.            

Results: Pretreatment with 300 mg/kg of omega3 fish oil supplements significant reduced the serum level of total cholesterol comparing with thioacetamide group. Pretreatment with the omega3 fish oil supplements all doses increased the serum level of glucose comparing with thioacetamide group but there was no significant difference. Pretreatment with the omega3 fish oil supplements at all doses showed no significant difference in serum levels of LDL, HDL cholesterol and triglyceride comparing with thioacetamide group.

Conclusion: Therefore, the result show that the protective effects of omega3 fish oil supplements on disorder of lipid function induced by thioacetamide in male rats. The protective effects of omega3 fish oil supplements can be due to their antioxidant effects.

Abstract

Background: Thioacetamide is a liver toxin that causes centrilobular necrosis. In this study, the protective effect of aqueous extract of Glycyrrhiza glabra root against liver-related biochemical factors changes induced by thioacetamide in male rats was investigated.

Materials and Methods: 35 male rats were divided into 5 groups of 7 : control group; sham group: receiving a single dose of 150mg/kg thioacetamide intraperitoneally; experimental groups 1 and 2 and 3: they received the aqueous extract of Glycyrrhiza glabra root at the doses of 100,200,300mg/kg daily orally during 3 months respectively and then a single dose of thioacetamide at 150 mg/kg as intraperitoneal injection . The serum levels of albumin, bilirubin and total protein were measured.

Results: The mean of body weight in all experimental groups receiving aqueous extract of Glycyrrhiza glabra root and thioacetamide did not show significant changes compared to the group receiving thioacetamide. The mean levels of serum bilirubin in all experimental groups receiving aqueous extract of Glycyrrhiza glabra root and thioacetamide did not show significant changes compared to the group receiving thioacetamide. The mean of serum albumin concentration in all experimental groups receiving aqueous extract of Glycyrrhiza glabra root and thioacetamide decreased significantly compared to the group receiving thioacetamide. The mean of serum total protein concentration in experimental groups receiving aqueous extract of Glycyrrhiza glabra root and thioacetamide did not show significant changes compared to the group receiving thioacetamide( p<0. 05).

Conclusion: The results of this study showed that the aqueous extract of Glycyrrhiza glabra root had protective effects against liver-related biochemical factors changes induced by thioacetamide in male rats.

Abstract

Introduction: Today about 27.4 percent of female 15-44 years and 1 percent of female in fertility age are affected by infertility. Iodine is a rare element that is essential for the synthesis of thyroid hormones. Concentration of the thyroid hormones in blood under the influence of iodine intake and changes in thyroid hormones levels interact with reproductive system. Today, all the people of Iran consuming iodized salt regardless of iodine status in their body. In this study according to high prevalence of the infertility among young couples, iodized salt intake on fertility in male rats were investigated.

Materials and Methods: In this study 20 male and 20 female adult Wistar rats were used. Twenty male adult Wistar rats were randomly divided into 2 groups. Including the control group and treatment group that received iodine and female adult Wistar were fed with a regular diet. Five male rats from each group were killed at the end of the fourth weeks in order to evaluate the possible effect of iodized salt on sperm analysis and weight of testis. After a month, male and female rats were placed in pairs in separate cages and their offspring were investigated in terms of number, gender and health.

Results: The result of this study showed that the number of healthy offspring of treated male rats was significantly lower than the control group.

Conclusion: Due to the negative effect of excessive iodine intake on fertility rate, it is recommended to couples to perform functional tests of their thyroid glands before intake of iodized salts.

Abstract

Background: The liver has important role in the metabolism of chemical drugs and plasma protein synthesis. Abilizol is used in the treatment of schizophrenia and bipolar maniac disorders. In this study the effect of abilizol have been observed on serum levels of liver enzymes, total protein, albumin and liver tissue changes.
Materials and Methods: In this experimental study 50 adult male rats from wistar race were divided in to 5 groups of 10.The experimental groups which received 5, 10 and 15 mg/kg of abilizol drug orally for 28 days. The control group which received nothings and the sham group which received solvent. At the end of this Period, blood samples were taken from all groups. ALT (Alanine Transaminase), AST (Aspartate Transaminase), ALP (Alkaline phosphatase), total protein and albumin were measured. Also histological changes of liver studied.
Results: No significant differences were observed in levels of ALT , ALP and albumin enzymes between experimental and control groups. In experimental group received 15mg/kg abilizol drug showed a significant decrease in level of AST in compared with control group. In experimental group received 15mg/kg abilizol drug showed a significant decrease in level of total protein in compared with control group (p≤0.05). Density of liver tissue in all experimental groups received abilizol drug decrease significantly in compared with control group.
Conclusion: Results show that abilizol is a cause of liver parenchymal cell failure and decreases liver enzymes levels.

Abstract

Background: Thyroid is an important organ with special function in metabolism of drugs. Primidone is a drugs for treatment of some patients including epilepsy. The present study was conducted to evaluate the effect Primidone on thyroid hormones function including T3,T4 and TSH and histological changes of thyroid newborn male rats.
Material and Methods: In this experimental study, 40 newborn male rats wistar strain divided into 5 group of 8 .The newborns were taken from pregnant mothers divided to the control group, the sham group and three experimental groups that received 20, 40 and 60 mg/kg b.w. primidone in the pregnancy period for 21 days orally. In 25 days after parturition, the newborns weighted and T3,T4 and TSH hormone assay. The newborns thyroid tissue evaluated by H&E histological staining methods.
Results: Body weight and thyroid weight in different experimental groups show did not significant difference compared to the control group. Concentration of TSH and T3 hormones in different experimental groups show did not significant difference compared to the control group. Also, serum level of T4 hormone in experimental groups 2,3 show a significant decrease compared to the control group. In tissue tissue samples with increasing doses of the drug was observed more necrosis.
Conclusion: Primidone has destructive effects on Thyroid gland.

Abstract

Background: Taraxacum Officinale is a medical plant with antioxidant benefit effects. The aim of the present study is to evaluate the effect of Taraxacum Officinale alcoholic extract on serum thyroid hormone levels in adult male rat.
Material and Methods: In this experimental study,45 adult male wistar rats were approximate average weight body of 200-250 gr were divided in to 5 groups of 9. Control ,sham ,and three experimental groups which received 50,100,150mg/kg of Taraxacum Officinale alcoholic extract by used gavage method. After this period (14 days) blood sample were collected and after measuring the amount of thyroid hormones of T3,T4,TSH hormones, data were analyzed by using SPSS software , ANOVA and t-test.
Results: The results showed that body weight average and serum level of T4 hormone in receiving maximum dose of Taraxacum Officinale alcoholic extract showed significant decrease comparing to control group. T3 serum level in experimental groups receiving Taraxacum Officinale alcoholic extract didn't show any significant difference comparing to control group. TSH serum level in experimental groups receiving Taraxacum Officinale alcoholic extract showed significant decrease comparing to control group (p<0.05).
Conclusion: The Taraxacum Officinale alcoholic extract reduced serum thyroid hormone levels in adult male rat.

At the end of the experiment , body weight were measured in all groups and then the blood sample was taken from heart and serum levels of AST, ALT, ALP and GGT enzymes were measured. Furthermore, The pathological changes of the livers were studied after hematoxylin-eosin staining. The results were analyzed using ANOVA and T-test statistical analysis.
Results: the results show that body weight in experimental groups had not a significant difference compared to control group. Serum level in AST, ALT, ALP, GGT in experimental groups had a significant increase compared to control group (p≤0/05). In tissue samples in experimental groups which were provided a higher level of necrosis was observed.
Conclusion: Primidone causes increase serum levels of AST, ALT, ALP, GGT and liver necrosis.

Abstract

Background: Primidone is effective in the treatment of epilepsy. In this research the effects of primidone on LH, FSH, testosterone hormones, body weight and testicular tissue in adult male rats, was studied.
Material and Methods: In this experimental study, 50 adult male Wistar rats, weighing approximately 180-200 g were divided into 5 groups of 10 animals. Groups: control group, Sham group and treatment at doses of 20, 40, 60 mg/kg, orally for 42 days (every other day) respectively. At the end, after determining the animal's body weight, blood samples was collected from the heart. Hormone concentrations of LH, FSH and testosterone were measured by RIA.Their testis were removed and weighed to evaluate the histological changes after hematoxylin-eosin staining.
Results: The body weight, testis weight and leydig and sertoli cells in all experimental groups showed no significant changes compared to the control group. The plasma levels of LH and FSH in all experimental groups increase significantly compared to the control group. The plasma level of testosterone Hormone and number of  spermatogonia,primary spermatocyte and spermatid in all experimental groups decrease significantly compared to the control group (P<0.05).
Conclusion:: primidone reduced levels of testosterone and increase levels of LH and FSH hormones.

Abstract

Background: Cadmium chloride causes liver dysfunction. In the present study, the protective effects of hydroalcoholic extract of aerial parts of Alcea rosea against changes in biochemical factors and hepatic tissue induced by cadmium chloride in male Males were investigated.

Methods: In this experimental study, 56 adult male Wistar rats weighing approximately 200±10 g  were divided into 6 groups of 9. The control group, The sham group 1 received 0.2 ml distilled water as solvent. The sham group 2 received 2 mg/ kg of cadmium chloride intraperitoneally for 21 days. Experimental groups 1, 2 and 3: 2 mg / kg cadmium chloride intraperitoneally for 21 days and then 450 and 300,150 mg / kg hydroalcoholic extracts of aerial parts of Alcea rosea received intraperitoneally for 30 days. At the end of the experiment, the body weight and liver weight of the animals were measured. Blood samples were taken to measure albumin, bilirubin and total protein levels. Liver tissue samples were studied pathologically after hematoxylin-eosin staining.

Results: The mean body weight in all experimental groups did not show significant changes compared to the sham group2. The mean liver weight in all experimental groups showed a significant increase compared to the sham group2. Mean serum bilirubin concentration in all experimental groups showed a significant decrease compared to the sham group2. The mean serum albumin concentration in the experimental group receiving 3 showed a significant increase compared to the sham group2. Mean serum albumin and total protein concentrations in the experimental group receiving 2 showed a significant increase compared to the sham group2 (P <0.05). In all experimental groups, the improvement of hepatic tissue changes induced by cadmium chloride was dose-dependent.

Conclusion: It is possible that the hydroalcoholic extract of the aerial parts of Alcea rosea improves the changes in biochemical factors and hepatic tissue induced by cadmium chloride in male rats.

Abstract

Background: Cadmium chloride causes  lipid dysfunction. In this study, the protective effects of hydroalcoholic extract of Passiflora caerulea aerial parts against lipid dysfunction induced by cadmium chloride in male rats were investigated.
Methods: In this experimental study, 54 adult male wistar rats were divided into 6 groups of 9. The control group, The sham group received  2 ml / distilled water as solvent. The negative control group  received 2 mg / kg cadmium chloride intraperitoneally for 21 days. Experimental 1, 2 and 3 groups: respectively, received 2 mg / kg cadmium chloride intraperitoneally for 21 days and then 150,300 and 450 mg / kg hydroalcoholic extracts of aerial parts of Passiflora caerulea intraperitoneally for 30 days. Blood samples were taken from all animals at the end of the experiment. Blood samples were taken to measure levels of HDL, LDL, total cholesterol and triglycerides. Data were analyzed according to SPSS18 program, analysis of variance (ANOVA) and Tukey tests.

Results: The mean serum concentrations of total cholesterol, triglyceride and low-density lipoprotein (LDL) in all experimental groups compared to the negative control group showed a significant decrease. Mean serum high-density lipoproteins (HDL) concentration in experimental 2 and 3 groups  showed a significant increase compared to the negative control group (P <0.05).

Conclusion: The hydroalcoholic extract of the aerial parts of Passiflora caerulea probably corrects the lipid dysfunction induced by cadmium chloride in male rats.

Abstract

Background: Curcumin has antioxidant and anti-inflammatory activities. The aim of this study was to investigate the protective effect of curcumin on changes in the serum concentration of LH, FSH , testosterone hormones and testicular tissue induced by methotrexate in adult male rats.

Materials and Methods: In this experimental study, 30 adult male Wistar rats with an approximate weight of 230-260 grams were divided into 6 groups of 5 each. Control group, sham group: 1 ml of distilled water as a solvent, experimental group 1: methotrexate (5 mg/kg), experimental group 2: curcumin (400 mg/kg), experimental group 3: methotrexate (5 mg/kg) + curcumin (200 mg/kg), experimental group 4 : methotrexate (5 mg/kg) + curcumin (400 mg/kg). After 28 days of treatment in the form of gavage injection, blood was taken directly from the heart to measure the serum concentration of  LH, FSH and testosterone hormones. The testicles were removed and tissue changes were examined after hematoxylin-eosin staining. The data results were analyzed based on one-way analysis of variance (ANOVA) and Tukey's test.

Results: In the group receiving methotrexate, the level of testosterone hormone, the number of spermatogonia, spermatocytes, sperm and leydig decreased and the serum level of LH and FSH increased significantly compared to the control and sham groups. In the groups receiving methotrexate + curcumin, curcumin was able to compensate the adverse effects caused by methotrexat (P <0.05).

Conclusion: Curcumin can improve the changes in serum testosterone, FSH, LH concentration and testicular tissue induced by methotrexate in adult male rats.

Abstract

Background: Saffron has anti-seizure, anti-depressant, anti-inflammatory, anti-tumor and memory improvement drugs. In this study, the effect of saffron hydroalcoholic extract on changes in liver-related biochemical factors following chronic vitamin A intoxication in adult male rats was investigated.






Methods: For this purpose, 48 adult male Wistar rats weighing approximately 200±20 g were divided into 6 groups of 8. Control group: did not receive any drug treatment, sham group: received only solvent extract, experimental groups 1: Hydroalcoholic extract of saffron alone received 50 mg / kg for 15 days. Experimental group 2: Vitamin A received 50,000 IU for 15 days. Experimental groups 3 and 4: First, they received 50,000 IU of vitamin A for 15 days and then received saffron hydroalcoholic extract at 50 and 80 mg / kg, respectively, for 21 days. At the end of the experiment, the body weight of the animals was measured. Blood samples were taken from the heart and albumin, bilirubin and total protein levels were measured. The results were analyzed using ANOVA and Tukey test.














Results: The mean body weight in the groups receiving vitamin A and saffron hydroalcoholic extract together and alone did not show a significant change compared to the control and sham groups. Mean serum levels of total protein and albumin in experimental group 2 receiving vitamin A and experimental group 3 showed a significant decrease compared to control and control groups. The mean serum levels of total protein and albumin in experimental group 4 showed a significant increase compared to experimental group 2 receiving vitamin A. The mean serum level of total bilirubin in experimental group 2 receiving vitamin A and experimental group 3 showed a significant increase compared to the control and sham groups. The mean serum level of total bilirubin in experimental group 4 showed a significant decrease compared to experimental group 2 receiving vitamin A (P> 0.05).
Conclusion: Saffron hydroalcoholic extract has a protective effect on changes in liver-related biochemical factors following chronic vitamin A poisoning in adult male rats.
